摘要

几乎所有的肿瘤都存在转化生长因子-β (transforming growth factor-b, TGF-β) 负反馈信号通路的缺陷。基因调控的表观遗传学机制包括DNA甲基化,这不仅对于正常细胞功能是必需的,而且在肿瘤形成中也至关重要。最新的研究显示,TGF-β信号通路介导了肿瘤发生及发展。随着肿瘤TGF-β信号通路中许多关键问题包括TGF-β1的自我诱导和DNA甲基转移酶(DNA methyltransferases, DNMTs)的高表达被相继发现,提示DNA甲基化在肿瘤发生发展中起着重要作用。在这篇综述中,我们总结了大量在前列腺癌中TGF-β信号通路与DNA甲基化有关的文献。结果显示前列腺癌中几乎所有检测到的DNA甲基化基因均与TGF-β信号通路有着直接或间接的关系。这些为我们将来研究、预防和治疗前列腺癌提供了前进的方向。
